The substrate is not fluorescent because the closely located nitrophenyl group quenches the fluorescence of the tryptophyl residue (2), but the hydrolytic product, tryptophylglycine is not constrained to remain in the vicinity of the nitrophenyl group and its fluorescence is not quenched.
