A significant increase of PTEN levels was found in NB4, HL-60 and THP-1 cells upon ATRA combined with EGCG treatment, paralleled by increased myeloid differentiation marker CD11b.
EGCG in synergy with ATRA promote degradation of PML/RARα and restores PML expression, and increase the level of nuclear PTEN.
These results therefore indicate that EGCG targets PML/RARα oncoprotein for degradation and potentiates differentiation of promyelocytic leukemia cells in combination with ATRA via PTEN.

In summary, it was found that (i) DAPK2 is essential for EGCG‐induced cell death in AML cells, (ii) ATRA and EGCG cotreatment significantly boosted neutrophil differentiation, and 67LR expression correlates with susceptibility of AML cells to EGCG.
Moreover, combined ATRA and EGCG treatment resulted in cooperative DAPK2 induction and potentiated differentiation.

Conclusions/Significance Our findings provide a molecular basis for the combination effect seen with dietary components, and indicate that ATRA may be a beneficial food component for cancer prevention when combined with EGCG.
In addition, the combined EGCG and ATRA treatment significantly suppressed the melanoma tumor growth in mice.

…,EGCG can block the TP-induced expression of the downstream … TLR4, including MyD88, NF[Formula: see text]B, and retinoid related orphan receptor (ROR-[Formula: see text]t), while EGCG can restore the TP inhibition …/winged-helix family transcriptional repressor p3 (FoxP3) that is the downstream target of Tim3.

In addition, EGCG increased expression of neutrophil differentiation markers such as CD11b, CD14, CD15 and CD66 in NB4 cells; and the combination of all-trans retinoic acid (ATRA) plus EGCG yield higher increase the expression of CD15 marker.

The results showed that there was an additive effect in leukemic cells when treated with a combination of ATRA and EGCG.
Thus, it was concluded that the cytotoxic effects of EGCG were improved by ATRA.
