Possible Interaction: Leucine and Melphalan

supplement:

Leucine

Research Papers that Mention the Interaction

Since L-leucine has been shown to be the most potent inhibitor of melphalan transport into cells in vitro, this amino acid was chosen for the present study in patients.
The area under the curve for melphalan was lower in seven of the patients after L-leucine.
The results indicate that L-leucine can reduce plasma melphalan levels in some patients, probably through a reduction in absorption of the drug from the gastrointestinal tract.

In all cells studied, L amino acids (1 mM) such as leucine , glutamine, tyrosine, and methionine reduced the cell-to-medium ratio of melphalan at 3 and 35 min.

The anti-cancer agent melphalan inhibited L-leucine uptake by MDA-MB-231 cells but had no effect on L-leucine efflux.

Acceleration of melphalan exodus by leucine is antagonized by α-amino-γ-guanidinobutyric acid, resulting in increased cellular retention of melphalan.

Kinetic analysis revealed that melphalan and leucine compete for transport carrier sites.

L-alpha-Amino-gamma-guanidinobutyric acid, the lower homolog of arginine, promotes melphalan uptake in vitro only in the presence of leucine.
Leucine completely abolishes this long-term survival by interfering with melphalan uptake into the tumor cells.
When administered to mice with melphalan and a dose of leucine which negates the 50% cure rate of melphalan, it reduces the therapeutic interference of leucine.

Deletion … leucine from RPMI 1630 medium increased the cytotoxicity produced … 35 min exposure of L1210 cells to melphalan by 10-fold and 2-fold, respectively, indicating that a substantial proportion of the protection afforded … from melphalan cytotoxicity in an amino acid environment can be attributed to these amino acids.

The LD90 for melphalan in the presence of 0.1 mM L-glutamine or L-leucine was increased by 7.3- and 10.8-fold respectively, under conditions where the cells had been pre-incubated with the amino acids.
