Possible Interaction: Gonadotropin-Releasing Hormone and Melatonin

Research Papers that Mention the Interaction

In the follicular phase, melatonin administration enhanced the LH and FSH responses to all three GnRH stimuli, whereas in the luteal phase, melatonin administration was ineffective.
In this study we investigated whether an increased pituitary response to GnRH is involved in the stimulatory effect of melatonin.
The present data indicate that an enhancing effect of melatonin on the LH and FSH responses to submaximal GnRH stimuli is evident in the follicular, but not the luteal, phase of the menstrual cycle and infer an endocrine window for the effect of melatonin on gonadotropin secretion.

Melatonin also has the capability to suppress the release of hypothalamic gonadotropin-releasing hormone (GnRH), a hormone that promotes cholangiocyte proliferation when serum levels are elevated.
Melatonin infusion reduced hypothalamic GnRH mRNA expression and led to decreased GnRH and increased melatonin secretion from cholangiocytes.

Melatonin can, therefore, influence the gonadal function indirectly--via its effect on gonadotropin-releasing hormone and/or gonadotropins secretion.

During the development, such tonic inhibitory effects of melatonin on GnRH action gradually decline due to a decrease in expression of functional melatonin receptors.
Their activation by melatonin leads to a decrease in cAMP production and activity of protein kinase A, and attenuation of gonadotropin-releasing hormone (GnRH)-induced gonadotropin secretion.

There have been good results in recent research to inhibit the generator of pulsatile secretion of GnRH necessary for ovulation by using melatonin in a new type of oral contraceptive.

Melatonin appears also to block GnRH secretion at the time of puberty.

The inhibitory action of melatonin is most likely exerted at the hypothalamic level, possibly through interference with the control of pulsatile secretion of gonadotropin-releasing hormone.

In addition, melatonin inhibits the secretion of gonadotropin-releasing hormone and may directly affect steroid metabolism within the ovaries and progesterone synthesis.
Melatonin is also closely related to changes in puberty, possibly because of its tonic inhibition of secretion of gonadotropin-releasing hormone during childhood.

Both GnRH and GnRH receptor mRNA levels were significantly decreased, to 61% and 45% of control levels, respectively, after melatonin treatment.
