Possible Interaction: Fluorouracil and Melatonin

supplement:

Melatonin

Research Papers that Mention the Interaction

Conclusions Melatonin exerts the effects of pro-apoptosis and anti-proliferation on gastrointestinal carcinomas, and might increase the sensitivity of 5-FU in GC and CRC patients.
Our results further showed that melatonin can downregulate the expression levels of 5-FU resistance-related genes, such as thymidylate synthase in GC and ATR , CHEK1 , BAX and MYC in CRC.
The qRT-PCR results demonstrated that melatonin enhanced the sensitivity of CRC 5-FU resistant cells by decreasing the expression of ATR .

Furthermore, a combination of melatonin and 5-FU markedly enhanced 5-FU-mediated cytotoxicity in 5-FU resistant cells.
RESULTS We observed that me atonin in hibited the cell growth in 5-F U re sistant CRC cells.

Furthermore, 1 mM melatonin significantly (P < 0.05) increased the cytotoxic effects of 5-FU.
Likewise, simultaneous stimulation with 1 mM melatonin and 1 mM 5-FU significantly (P < 0.05) enhanced the ratio of cells with an overproduction of intracellular reactive oxygen species and substantially augmented the population of apoptotic cells compared to the treatment with 5-FU alone.
Nonetheless, melatonin only displayed moderate chemosensitizing effects in CIS-treated HT-29 cells, as suggested by a slight increment in the fraction … that was observed only after 48 h. Consistently, co-stimulation of HT-29 cells with 20 µM CIS or 1 mM 5-FU in … mM melatonin further ….
Overall, our results indicate that melatonin increases the sensitivity of HT-29 cells to 5-FU treatment and, consequently, the indolamine could be potentially applied to colorectal adenocarcinoma treatment as a potent chemosensitizing agent.

Importantly, melatonin effectively enhanced cytotoxicity of 5-Fu to ESCC in vitro and in vivo.

By contrast, the inhibitory effect of melatonin was reduced by the co-administration of 5-FU , independently of 5-FU concentration.

However, co-treatment with melatonin ameliorated the reductions in neurogenesis caused by 5-FU.

The positive effect of melatonin was also seen in the rats that received 5-FU.

The synergistic effects of the combined treatment were evident in many other cancer cell lines, and melatonin was also highly effective in inducing cancer death when combined with other cancer drugs, including cisplatin, 5-fluorouracil , irinotecan, and sorafenib.

Combination of 5-FU with melatonin significantly decreased the IC50 value of 5-FU from 100 μM to 50 μM. Moreover, combination therapy increased intracellular levels of ROS and suppressed antioxidant enzymatic activities (P < 0.05).
XIAP and survivin expression levels potently decreased after combination treatment with melatonin and 5-FU (P < 0.05).

Moreover, melatonin restored antioxidant enzyme levels and reduced oxidative stress in the hippocampus and PFC caused by 5-FU.
The results showed that melatonin reduced the number of p21-positive cells in the SGZ of the dentate gyrus and increased Nrf2, DCX and BDNF protein expression in rats treated with 5-FU.
These findings reveal a mechanism of the neuroprotective properties of melatonin against 5-FU in a rat hippocampus and PFC.
