Possible Interaction: Ethanol and Midazolam

supplement:

Ethanol

Research Papers that Mention the Interaction

One hour following treatment with midazolam and midazolam + alcohol , critical flicker fusion threshold (CFFT) was significantly depressed and subjects perceived themselves as feeling more sedated when compared to treatment with placebo or alcohol alone.
Stimulus processing time was significantly increased at one hour after treatment by midazolam taken in conjunction with alcohol , thus resulting in an overall increase in total reaction time.

Acute doses of midazolam , alcohol and midazolam in combination with alcohol were all found to decrease iconic memory whereas only midazolam and midazolam + alcohol impaired immediate recall.
Anterograde amnesia was observed following treatment with midazolam and midazolam in conjunction with alcohol.
The results suggest a potentiation of the effects of midazolam , on aspects of human memory by the co-administration of small doses of alcohol.

Midazolam increased ethanol drinking, as expected, when administered on successive days.

In the presence of ethanol the values of the EC50 of midazolam and zolpidem were reduced to approximately 50% of the original value.

Among the other drugs tested, only midazolam produced substantial substitution for ethanol.

ICV injections of AP and midazolam shortened the latency and prolonged the duration of sleep induced by IP injections of ethanol (5.0 g/kg).

In the first experiment, it was found that midazolam (5 mg/kg) increased home cage ethanol drinking 0–2 h after administration.

These findings show that there is a pharmacodynamic interaction between midazolam and ethanol in vivo.

The intraperitoneal … of midazolam produced statistically significant decreases in levels of DOPAC and ratio of … values (P < 0.01) in rats 48 hr after withdrawal of ethanol , with respect to control and chronically ethanol-treated animals, in contrast to the absence of changes produced when injecting thiopental or somatostatin.

An infusion of β-CCT given alone increased CMRo2 alone and reversed the depression in both cortical CBF and CMRo2 produced by midazolam plus alcohol.
Midazolam infusions alone decreased cortical CBF and CMRo2, 35–40%, while 2.5 mg/g alcohol (which did not depress CMRo2, alone) combined with midazolam produced a 70% depression of cortical CBF and metabolism.
