Possible Interaction: Ethanol and Mianserin

supplement:

Ethanol

Research Papers that Mention the Interaction

Mianserin and alcohol impaired objective performance in most tests, and their combination showed at least an additive effect.

Considering these antecedents, we explored modulation of the acute stimulating effect of ethanol (2.5g/kg) by two anxiolytic drugs, Mianserin (2.5 or 5mg/kg) and Ondansetron (1 or 3mg/kg).
Mianserin attenuated the stimulating effect of ethanol at a dose that did not affect locomotor activity in water-controls, likely acting through 5-HT2 receptors, while Ondansetron, a 5-HT3 antagonist, did not affect this response.

Desipramine, mianserin and alaproclate also increased ethanol plasma levels initially, but to a lesser extent.

In contrast, both single dose and repeated administration of mianserin prevents increased locomotor activity of animals observed after ethanol (2.5 mgkg(-1)).
Mianserin administered as a single dose of 5 mgkg(-1) was found to decrease LD(50) of ethanol and its ED(50) in rota-rod test.
Mianserin administered as a single dose of 5 or 20 mgkg(-1) prolongs ethanol sleeping time in mice but given daily for 14 days has no influence on this time.

However, when administered concurrently with ethanol treatment, mianserin significantly antagonized the reductions in cortical GRE-DNA binding and in GR protein expression in cortical, PVN, and central, but not medial and basolateral, amygdaloid structures during ethanol withdrawal.

Mianserin , injected as a single dose given either 1 hr (0.16-5 mg/kg, ip) before testing or given (20 mg/kg, … of the 3rd day of ethanol administration, i.e., 48 hr and 7 days before testing, dose-dependently prevented or reversed the ethanol withdrawal induced … activity.

During conditioning, ethanol produced increases in locomotor activity that were reduced by mianserin.
However, mianserin enhanced the acquisition of ethanol preference, whereas mianserin alone did not produce either place preference or aversion.
