Possible Interaction: Desipramine and Epinephrine

supplement:

Epinephrine

Research Papers that Mention the Interaction

After desipramine , cardiac responsiveness to epinephrine was markedly enhanced, although more (P < 0.01) in young vs. older subjects for heart rate and cardiac index (+14 vs. 7 beats/min and +1.6 vs. 1.1 l.min(-1).m(-2), respectively, at 20 ng.kg(-1).min(-1)).
Combined with desipramine and trimetaphan, cardiac responses to epinephrine were further enhanced, again more (P < 0.01) in young subjects, resulting in large differences in heart rate and ejection fraction increases (+29 vs. 17 beats/min and +14 vs. 7%, respectively, at 20 ng.kg(-1).min(-1)).

After desipramine , cardiac extraction of epinephrine was reduced to 12 +/- 2% and [3H]norepinephrine to 20 +/- 3%.
In rabbits desipramine reduced the plasma clearance of [3H] epinephrine by 18%, significantly less than the 42% reduction in [3H]norepinephrine clearance; production of DHPG from epinephrine was less than half the production of [3H]DHPG from [3H]norepinephrine.
In subjects infused simultaneously with [… and [3H] norepinephrine, desipramine reduced the … of [3H]epinephrine by 28 +/- 6%, significantly less than the 49 +/- 7% reduction in [3H]epinephrine extraction; the plasma clearance of […]epinephrine was reduced by 4 +/- 5%, significantly less than the 20 +/- 6% reduction in [3H]norepinephrine clearance.

Desipramine , however, reduced the mixed venous baseline plasma levels of noradrenaline (47%) and DOPEG (40%), and tended to reduce that of adrenaline (34%).

desipramine (blockade of neuronal uptake) in control subjects … response: ΔFS during 40 ng/kg · min epinephrine amounted to 17.6±3.6% (p<0.001 versus untreated controls, NS versus heart transplant recipients) …±0.8 mm Hg/mm (p<0.001 versus untreated controls, NS versus heart transplant recipients).

Intravenous administration of desipramine (1 mg kg−1) significantly increased plasma NE concentration but decreased plasma Epi concentration in both groups, suggesting that neuronal NE uptake had contributed to the reduction in plasma NE concentration.

After desipramine , cardiac spillovers of adrenaline were decreased, whereas those of noradrenaline were increased so that the ratio of adrenaline to noradrenaline spillover, meaned before and during stimulation, decreased substantially from 1:42 to 1:166.
This reversed the situation before blockade so that desipramine substantially decreased the ratio of adrenaline to noradrenaline spillover.

Desipramine enhanced much more the postjunctional effect of noradrenaline, adrenaline , and α-methylnoradrenaline in the epididymal than in the prostatic portion of the rat vas deferens.
Moreover, desipramine similarly enhanced the effects of noradrenaline and adrenaline in the ear artery, while it did not modify the responses to these amines in the jejunal artery.

Desipramine decreased pulmonary … and epinephrine clearance and spillover to near-zero levels, whereas whole body norepinephrine clearance fell by 51 +/- 3% (P < 0.001), norepinephrine spillover by 54 +/- 6% (P < 0.005), epinephrine clearance by 30 +/- 6% (P < 0.01), and epinephrine spillover by 34 +/- 11% (P < 0.05).

Although the basal Epi concentration was undetectable, local administration of desipramine increased Epi concentration of the dialysate to 38.1+/-18.5 pg/ml.

Desipramine reduced the clearance of NE, Epi , and DA by 39, 13, and 14%, respectively, but did not affect Iso clearance.
