Possible Interaction: Cocaine and Gonadotropin-Releasing Hormone

Research Papers that Mention the Interaction

At 15 days, the GnRH IPI was significantly decreased after incubation with cocaine and amphetamine‐regulated transcript (CART), α‐melanocyte‐stimulating hormone, corticotrophin‐releasing factor (CRF) and neuropeptide Y (NPY).

On the basis of these results it is suggested that acutely administered cocaine stimulates release of hypothalamic GnRH , which, in turn, stimulates pituitary gonadotrophin secretion.

Cocaine stimulates a rapid release of gonadotropins in intact rhesus males and females and human males and enhances synthetic luteinizing hormone-releasing hormone (LHRH), stimulated luteinizing hormone (LH) and follicle-stimulating hormone (FSH).

Our results that cocaine and tetrodotoxin attenuated phorbol ester stimulation of GnRH , presumably through inhibition of Na+ influx, suggest a direct biochemical mechanism for cocaine disruption of hypothalamic GnRH secretion and, consequently, cyclic reproductive function.
PDBu-stimulated GnRH release was blocked by both tetrodotoxin and cocaine , known inhibitors of Na+ influx.

Cocaine (0.4 mg/kg) significantly enhanced LHRH stimulation of LH compared to placebo or 0.8 mg/kg cocaine administration (P less than 0.01).
LHRH stimulated a significant increase in LH within 10 min after placebo-cocaine administration (P less than 0.05) and after each dose of cocaine (P less than 0.0001).
