Possible Interaction: Cocaine and Ghrelin

supplement:

Cocaine

drug:

Ghrelin

Research Papers that Mention the Interaction

Overall, these findings provide further evidence that mesolimbic ghrelin signaling is indeed critically involved in mediating the rewarding effects of cocaine.

Combined subthreshold dosing of VTA ghrelin with a subthreshold dose of cocaine also evoked reliable increases in intake compared to vehicle.
Pretreatment with ghrelin potentiated the effect of cocaine on ethanol intake at a low dose of 2.5 nmol, whereas 10 nmol suppressed cocaine-induced ethanol intake.
To further investigate a possible interaction between these two neurochemical systems, we examined the impact of ghrelin, cocaine and combined injections of ghrelin with cocaine , on voluntary ethanol intake.

Ghrelin , an orexigenic hormone produced by the stomach, increases food intake and enhances the locomotor and rewarding effects of cocaine.

Rats pretreated with 5 nmol or 10 nmol ghrelin showed an enhanced locomotor response after treatment with 15 mg/kg cocaine relative to rats treated with 0 nmol ghrelin.
These results indicate that acute injection of ghrelin , at a feeding-relevant dose, can augment the acute effects of cocaine on locomotion in rats.

Moreover, systemically administered ghrelin crosses into the brain and is known to augment the locomotor-stimulating effects of cocaine [COC: Wellman et al., 2005].

Cocaine increased locomotion as a function of dose in the 0 nmol ghrelin group, but the effect of cocaine was even greater when preceded by 5 nmol ghrelin.
These results indicate that acute injection of ghrelin , at a feeding-relevant dose, augments the acute effects of cocaine on locomotion in rats.

In order to further investigate ghrelin’s role in dopamine-mediated reward, the present report examined whether pretreament with ghrelin , administered directly into the ventral tegmental area (VTA) of the midbrain, would potentiate the rewarding properties of cocaine as measured by CPP.
Our results indicated that both cocaine and ghrelin elicited CPP and that ghrelin pre-treatment potentiated the effect of cocaine on place preference.
