Possible Interaction: Cisplatin and Andrographolide

supplement:

Andrographolide

Research Papers that Mention the Interaction

Compared with the sole cisplatin treatment, combining cisplatin with Andro potentially inhibited tumor growth, reduced the incidence of lung metastases, and relieved renal tubular damage.
These results suggest that Andro offers an ideal candidate of autophagy inhibitors in clinical application, and combination of cisplatin with Andro could be a promising strategy for the treatment of lung cancer.

Andro potentiated the cytotoxic effect of CDDP in Lovo cells through apoptosis.
Finally, the combination therapy of Andro with CDDP was evidenced by its synergistic inhibition on the growth of Lovo cells in xenograft tumor studies.
The addition of Andro to CDDP induced synergistic apoptosis, which could be corroborated to the changes in protein and mRNA levels of Bax and Bcl-2, and the increased Fas/FasL association in these cells, resulting in increased release of cytochrome c, and activation of caspases.

Pretreatment of endothelial cells with andrographolide prior to stimulation with cisplatin resulted in activation of PI3K/Akt and eNOS, production of nitric oxide (NO) and cGMP, with a consequential lowering of endothelial cell leakiness and improved transendothelial electrical resistance.

Our data indicate that the combination treatment of andrographolide and DDP results in synergistic anti-tumor growth activity against oral squamous cell carcinoma CAL-27 in vitro and in vivo.
The results showed the combination of andrographolide and DDP synergistically inhibited CAL-27 cell proliferation in vitro and caused tumor regression in vivo in the CAL-27 xenografts.
These results demonstrated that combination of andrographolide with DDP was likely to represent a potential therapeutic strategy for oral squamous cell carcinoma.

Synergism was observed when Cis and Andro were administered using the sequences 0/4 h and 4/0 h. The percentage of apoptotic cell death was found to be greater for the 0/4 h combination of Andro and Cis as compared to those values from single-drug treatments.

Finally, curcumin and andrographolide increased cytotoxicity following co‐treatment with either cisplatin or doxorubicin, two chemotherapeutic agents widely used in the clinical management of NB.

AG might act as a chemosensitizer when co-administered with 5-FU, ADM and DDP , and the mechanism of reversal modulation of multidrug resistance by AG in the HCT-8/5-FU resistant cell line might be related to its downregulation of overexpression of P-170.
However, a remarkable inhibitory and apoptosis rate was shown when AG was co-administered with 5-FU, ADM and DDP , respectively.
