Possible Interaction: Amphetamine and Pimozide

supplement:

Amphetamine

Research Papers that Mention the Interaction

Like other neuroleptics, pimozide antagonises many of the actions of amphetamine and apomorphine, such as stereotyped behaviour and emesis, but it is not so effective as haloperidol or chlorpromazine in protecting animals against lethal doses of noradrenaline.
Single oral doses of pimozide , 5 to 20mg, reduced the subjective euphoric response to subsequent intravenous injections of amphetamine in abusers of the drug.

The antistereotypic abilities of haloperidol, fluphenazine and pimozide were most marked against amphetamine , but this was not a consistent observation for thioridazine, clozapine and sulpiride.

The latter action of amphetamine was blocked by pimozide.

Pimozide and lithium alter behavior and the behavioral effects of amphetamine , but neurotransmitters other than NE may be involved.

Single doses of clozapine, risperidone, and pimozide all markedly decreased calling under saline and AMPH.

DA blockade pimozide , 0.0625 to 0.5 mg/kg, or (+)-butaclamol, 0.0125 to 0.1 mg/kg) caused periods of increased rate of responding for amphetamine which were followed, in the case of higher doses, by response cessation.
When access to amphetamine injections was delayed until 4 h after animals received 0.5 mg/kg pimozide , rate of responding was elevated.

When amphetamine treatment was suspended in Phase IV, pimozide accelerated the extinction, especially in those groups that had previously received the 1.0 mg/kg dose of amphetamine.

Such activity seemed related to a catecholaminergic substrate since the increase of responding induced by amphetamine was blocked by pimozide , d,l-propranolol and prazosin.

Pimozide blocked amphetamine anorexia at 0.5 and 1.0 mg/kg, suggesting that at both doses amphetamine anorexia has a dopaminergic substrate.

The effects of pimozide, clonidine and amphetamine were approximately additive amphetamine cancelled the effects of pimozide and clonidine, while clonidine augmented the effect of pimozide).
