Possible Interaction: Agmatine and Clonidine

supplement:

Agmatine

Research Papers that Mention the Interaction

Clonidine synergized the effect of agmatine on avoidance response following nicotine withdrawal.
In separate experiments, &agr;2‐adrenoceptor agonist, clonidine (0.5–1&mgr;g/rat, icv) not only demonstrated significant increase in the step through latency as in nicotine withdrawn rats but also potentiated the pharmacological effect of agmatine.

The orexigenic effect of agmatine and GABAergic agonists was potentiated by a α2-adrenoceptors agonist, clonidine (10ng/rat, i.c.v.)

In combination studies, the anticompulsive-like effect of agmatine (10mg/kg, ip) was significantly potentiated by prior administration of moxonidine (0.25mg/kg, ip) or clonidine (30µg/kg,) or 2-BFI (5mg/kg, ip).

Both moxonidine and clonidine failed to affect carbachol or adenosine-triphosphate-induced contractions; however, 1000.0 µmol/L agmatine significantly increased these contractions.

Coadministration of agmatine (20 microg, icv) and alpha(2)-adrenoreceptors agonist, clonidine (0.1 microg, icv) evoked synergistic inhibition of nicotine sensitization.

Interestingly, the vasopressor responses to exogenous noradrenaline, which remained unaffected during the infusions of physiological saline, B-HT 933, moxonidine, cirazoline and agmatine , were significantly blocked during the infusions of clonidine or methoxamine.

Only the highest dose of agmatine (1000 µg/kg) did reverse the antihypertensive effects of rilmenidine and clonidine.
Yet agmatine produced a small tachycardia at relatively low doses and was able to reverse the bradycardia induced by clonidine.

Agmatine concentration‐dependently inhibited the binding of [3H]‐clonidine and [3H]‐rauwolscine to rat brain cortex membranes (Ki values 6 μM and 12 μM, respectively).
In addition, 30 and 100 μM agmatine increased the rate of association and decreased the rate of dissociation of [3H]‐clonidine resulting in an increased affinity of the radioligand for the α2D‐adrenoceptors.

Agmatine when administered at the highest well-tolerated dose of 10 micrograms/kg did not further alter the clonidine-induced hypotension but produced a greater bradycardia (-12 +/- 4 beats/min clonidine; -29 +/- 4 beats/min clonidine plus agmatine ; p < 0.05).
